| The rarity of testicular germ cell tumors, in conjunction with the young age of its patients, makes it challenging to study its long-term adverse effects. It is believed that the psychosocial state of testicular cancer patients is adversely impacted during treatment and survivorship. As such, Khalid Alkhatib and colleagues sought to investigate the changes in HADS scores over time in our institutional cohort of testicular cancer survivors, hypothesizing that anxiety and depression indices will improve over time, and specifically after treatment completion.

| PSMA has already been utilized as a target to generate a therapeutic response – with the approval of Pluvicto (based on the VISION trial data), PSMA targeted therapy has become an important adjunctive treatment in patients with mCRPC. However, PSMA may be targeted by antibodies (mAb) or small molecules (SML), with different kinetics and biodistribution. |

| In this presentation, Tanya Dorff discusses the final results from a phase I study of PSCA-targeted chimeric antigen receptor (CAR) T cells in patients with metastatic castration resistant prostate cancer (mCRPC). Based on this data, Dr. Dorff and colleagues report that PSCA-CAR T cells have anti-cancer activity in mCRPC with DLT of cystitis. PSCA is rest in mCRPC, with >80% screened patient having at least 2+ tissue expression. |

He emphasizes that PSCA appears to be a good target for metastatic castration resistant prostate cancer. Lymphodepletion chemotherapy process allows expansion and persistence of CAR-T cells but it can contribute to dose limiting toxicities such as cystitis. Safety is still a concern for CAR-T technology. It is not enough to have a good target and likely success depends on multiple factors.

FGFR3 alterations were seen in only 19% of all patients screened for PROOF-302, a trial that was enriched for upper tract urothelial carcinoma: 30% of upper tract urothelial carcinoma and 13% of urothelial carcinoma of the bladder. |
