| The analysis of the JAVELIN Bladder 100 trial, efficacy by duration or number of cycles of first-line chemotherapy presented at the 2021 ASCO GU helped clinicians understand the outcomes of avelumab from the JAVELIN 100 trial. Alicia Morgans and Cora Sternberg, co-investigator of the JAVELIN 100 trial highlight findings from this analysis showing all of the patients benefited from the avelumab therapy, compared to the patients who didn't receive avelumab.

| Yohann Loriot presents results of the post hoc analysis of the JAVELIN Bladder 100 trial, specifically efficacy by duration or number of cycles of first-line chemotherapy. Improved overall survival was observed with avelumab first-line maintenance versus best supportive care alone irrespective of duration or cycles of first-line chemotherapy received prior to entering the trial. Among patients who stopped first-line chemotherapy prior to 6 cycles, avelumab first-line maintenance still provided an overall survival benefit.

| In this presentation, Petros Grivas discusses the overall survival data within pre-specific patient subgroups from the JAVELIN Bladder 100 trial. The overall conclusion of the JAVELIN Bladder 100 trial that maintenance avelumab results in significantly longer overall survival in patients with advanced urothelial carcinoma who have stable disease after first-line platinum chemotherapy appears to be consistent across all studied subgroups.

| Platinum-based chemotherapy is standard-of-care first-line treatment for advanced urothelial carcinoma. However, progression-free survival and overall survival are limited by chemotherapy resistance. In a phase 3 trial, the authors randomly assigned patients with unresectable locally advanced or metastatic urothelial cancer who did not have disease progression with first-line chemotherapy (four to six cycles of gemcitabine plus cisplatin or carboplatin) to receive best supportive care with or without maintenance avelumab. |
