Matthew Galsky, MD
| First Results from the Phase 3 CheckMate 274 Trial of Adjuvant Nivolumab vs Placebo in Patients Who Underwent Radical Surgery for High-Risk Muscle-Invasive Urothelial Carcinoma were presented at ASCO GU 2021 and are the topic of discussion between Matthew Galsky and Alicia Morgans.

| Avelumab First-Line Maintenance Plus Best Supportive Care vs Best Supportive Care Alone For Advanced Urothelial Carcinoma: JAVELIN Bladder 100 Subgroup Analysis Based On Duration And Cycles Of First-Line Chemotherapy
| Yohann Loriot, MD, Ph.D.
| Yohann Loriot presented results of a post hoc analysis of the JAVELIN Bladder 100 trial, specifically efficacy by duration or number of cycles of first-line chemotherapy. Improved overall survival was observed with avelumab first-line maintenance versus best supportive care alone irrespective of duration or cycles of first-line chemotherapy received prior to entering the trial. Among patients who stopped first-line chemotherapy prior to 6 cycles, avelumab first-line maintenance still provided an overall survival benefit.

| First Results from the Phase 3 CheckMate 274 Trial of Adjuvant Nivolumab vs Placebo in Patients Who Underwent Radical Surgery for High-Risk Muscle-Invasive Urothelial Carcinoma
| Dean F. Bajorin, MD
| Dean Bajorin presents the results of CheckMate-274 assessing the role of adjuvant nivolumab in patients following radical resection with or without neoadjuvant chemotherapy with cisplatin. The data demonstrates that adjuvant nivolumab is associated with both statistically significant and clinically meaningful improvement in disease-free survival compared to placebo in patients with muscle invasive urothelial carcinoma following radical surgery, both in ITT patients and patients with PD-L1 ≥ 1%.

| Atezolizumab Monotherapy vs Chemotherapy in Previously Untreated Locally Advanced or Metastatic Urothelial Carcinoma: Clinical Outcomes by PD-L1 Status In Cisplatin-Ineligible Patients From the Phase III IMvigor130 Study
| Matthew Galsky, MD
| Matthew Galsky presents on the results of the exploratory analysis of IMvigor130 assessing outcomes by PD-L1 status in cisplatin-ineligible patients. In this exploratory study, results suggested improved overall survival with atezolizumab monotherapy versus placebo plus platinum/gemcitabine in cisplatin-ineligible patients with IC2/3 status, and objective response rates were higher in with atezolizumab in this subgroup.

| Primary results of EV-301: A Phase III Trial of Enfortumab Vedotin vs Chemotherapy in Patients With Previously Treated Locally Advanced or Metastatic Urothelial Carcinoma
| Thomas Powles, MD, MBBS, MRCP
| Thomas Powles presented results of EV-301, a phase III trial of enfortumab vedotin versus chemotherapy in patients with previously treated locally advanced or metastatic urothelial carcinoma. Enfortumab vedotin had superior overall survival and progression-free-survival compared with chemotherapy. Enfortumab vedotin also demonstrated a tolerable and manageable safety profile, with no new safety signals identified.

| TIVO-3: Tivozanib in Patients with Advanced Renal Cell Carcinoma Who Have Progressed After Treatment with Axitinib |
| Brian I. Rini, MD |
| Brian Rini presented results of the TIVO-3 trial testing tivozanib in patients with advanced RCC who had progressed after prior axitinib treatment. The results suggest that a selective VEGFR inhibitor, tivozanib, is active after prior axitinib and provides superior benefit compared to the multi-targeted VEGFR-TKI sorafenib. |

| Sia Daneshmand, MD
| Sia Daneshmand discussed the SEMS (Surgery in Early Metastatic Seminoma) trial, a phase 2 study of RPLND as first line treatment for testicular seminoma with isolated retroperitoneal disease measuring 1-3 cm. the SEMS trial establishes additional data for RPLND as a treatment alternative for testicular seminoma with isolated retroperitoneal lymphadenopathy (1-3 cm).

| Imaging Modality and Frequency In Surveillance of Stage I Seminoma Testicular Cancer: Results From A Randomized, Phase III, Factorial Trial (TRISST)
| Robert Huddart, MB, BS, MRCP, FRCR, Ph.D.
| Surveillance for recurrence consists of history and physical exam, tumor marker profiling, and intermittent imaging with CT scans. However, the evidence supporting any particular surveillance strategy is quite limited. Furthermore, the many CT scans typically utilized for surveillance may cause unnecessary radiation exposure. To determine if the number of CT scans utilized for surveillance could be decreased or replaced with MRI, the randomized phase 3 trial TRISST was designed.
