The ARASENS trial is an international, double-blind, phase 3 study that enrolled patients with metastatic hormone-sensitive prostate cancer (mHSPC) and ECOG performance status 0 or 1. The most important finding was androgen deprivation therapy (ADT) plus docetaxel plus darolutamide produced a better overall survival than ADT plus docetaxel and placebo. |

| The main question is whether the benefit in terms of OS and PFS seen between triplet therapy vs. docetaxel plus ADT holds true also vs. AAB plus ADT. In other words, is docetaxel needed? To disentangle this question, the authors conducted a systematic review and meta-analysis on chemotherapy and AAB alone or combined for men with mHSPC. To do so, we relied on survival data reconstruction. We were able to digitalize the Kaplan-Meier curves for PFS and OS and to extrapolate quasi-individual survival data, i.e. for each individual the time from randomization to either censoring or event and the event of censoring itself.

| David Crawford presented a post hoc analysis of safety and adverse events among men with metastatic hormone-sensitive prostate cancer (mHSPC) treated in the context of the ARASENS trial. ARASENS demonstrated that the triplet combination of darolutamide+androgen-deprivation therapy (ADT)+docetaxel significantly reduced risk of death by 32.5% (hazard ratio [HR] 0.675; 95% confidence interval [CI] 0.568–0.801; P <0.0001) compared to the doublet of ADT+docetaxel.
