Dr. Carroll was a urologic oncology fellow under Dr. Whitmore at Memorial Sloan Kettering Cancer Center (MSKCC) in 1986 and throughout his talk provided several lessons learned, not just from his time at MSKCC, but also throughout his career.

| Fred Saad presents an exploratory analysis of the efficacy of olaparib versus either abiraterone or enzalutamide, given a potential difference in the efficacy of NHA sequencing. Dr. Saad concluded that this exploratory analysis of the PROfound trial demonstrates improved outcomes of olaparib compared to both abiraterone and enzalutamide, without meaningful differences in outcomes in the control arm on the basis of which NHA was used. Further, he emphasized that sequential use of NHA appears to have limited benefit. |

| Prostate-Specific Antigen Kinetics in Patients With Advanced Prostate Cancer Treated With Apalutamide: Results from the TITAN and SPARTAN Studies

| Kim Chi presented a post hoc analysis of prostate-specific antigen (PSA) kinetics in patients enrolled in the TITAN and SPARTAN trials. Patients with advanced PC, whether mCSPC or nmCRPC, who received apalutamide +ADT demonstrated rapid PSA declines that continued over time. There was a high rate of patients with PSA90 and with PSA≤0.2 ng/mL responses, with a majority of patients reaching PSA90 by 12 months. Those patients who had PSA90 and/or PSA nadir of ≤0.2 ng/mL had improved long-term oncologic outcomes.

| Alicia Morgans explored the role of systemic treatment intensification in metastatic castration sensitive prostate cancer (mCPSC). She concludes that treatment intensification with either docetaxel or an androgen receptor targeting agent is the standard of care for patients with mCSPC. The choice among these agents depends on quality of life and patient preferences in the context of shared decision-making. |

| Practical Clinical Applications for AR Inhibitor Therapy |

| To close the plenary session entitled “Advances in Prostate Cancer: Androgen Deprivation Therapy Across the Disease Continuum” held in conjunction with the American Urologic Association Annual Meeting this evening, Dr. Dan Lin hosted a discussion of pertinent clinical questions along with Dr. Alicia Morgans and Dr. David Penson. In this session, Dr. Lin had laid out the landscape of nmCRPC and mCSPC while Drs. Penson and Morgans highlighted novel treatment approaches for each of these disease states, respectively. |

| In this presentation Kelvin Moses, addresses controversies in the management of high-risk prostate cancer. Dr. Moses emphasized that high-risk disease represents 15% of patients diagnosed with localized prostate cancer and early identification of high-risk characteristics and the extent of disease may allow for tailored therapy. However, most will benefit from a multi-modal approach.

| Ronald Chen addressed the question of designing the ideal randomized controlled trial in high-risk localized prostate cancer at the American Urological Association's 2021 Annual meeting. Dr. Chen emphasized the need for high-quality data comparing radical prostate and radiotherapy in high-risk prostate cancer. Carefully designed randomized controlled trials and non-randomized prospective studies can provide important high-quality evidence. |
