| Biomarker-Directed Therapy in Black and White Men With Metastatic Castration-Resistant Prostate Cancer
|
| Clara Hwang, MD
|
| Clara Hwang presented an analysis of biomarker-directed therapy in men with metastatic castration-resistant prostate cancer (mCRPC). In a real-world mCRPC molecular profiling cohort, there are similar overall rates of actionable molecular alterations in non-Hispanic black and non-Hispanic white men, but higher rates of MMRd/MSI-H and lower frequency of PTEN alterations in non-Hispanic black men. Further, there was not evidence of differences in the delivery of targeted therapy.

| Racial Concordance and Trust in Health Communications: A Randomized Trial of Videos About Prostate Cancer
|
| Stacy Loeb, MD
|
| Stacy Loeb presented a randomized trial of 2904 U.S. adults age ≥40 that sought to evaluate the association between racial representation in online content about prostate cancer and trust in the content. A secondary objective was to identify additional attributes that influence trust in online content. While there is no difference among White adults, there is a significant association between racial concordance of the presenter and trust in prostate cancer information among Black adults.

| Scott T. Tagawa, MD |
| Scott Tagawa discussed the tolerability of theranostic treatment with 177Lu-PSMA among men with metastatic castration-resistant prostate cancer (mCRPC), stratified by their treatment exposure, in men treated within the VISION trial. |

| Health-Related Quality of Life and Pain in the MAGNITUDE Study of Niraparib With Abiraterone Acetate and Prednisone in Patients With Metastatic Castration-Resistant Prostate Cancer and HRR Gene Alterations |
| Dana E. Rathkopf, MD |
| Dana Rathkopf discussed health-related quality of life (HRQoL) outcomes among patients treated with niraparib and abiraterone in men with previously untreated metastatic castration-resistant prostate cancer (mCRPC) treated in the MAGNITUDE trial. |

| Overall Survival and Biomarker Results From Combat: A Phase 2 Study of Bipolar Androgen Therapy Plus Nivolumab for Patients With Metastatic Castrate-Resistant Prostate Cancer |
| Mark Markowski, MD, PhD |
| Mark Markowski discussed bipolar androgen therapy (BAT) in patients with metastatic castrate-resistant prostate cancer (mCRPC). There are favourable outcomes of the combination of BAT and nivolumab in heavily pretreated patients with mCRPC, with a median overall survival of exceeding 2 years and durable responses in a subset of patients. |

| DASL-HiCaP: Darolutamide Augments Standard Therapy for Localized Very High-Risk Cancer of the Prostate (ANZUP1801)—A Randomized Phase 3, Double-Blind, Placebo-Controlled Trial of Adding Darolutamide to ADT and Definitive |
| Tamim Niazi, MD |
Tamim Niazi described the rationale, design, and protocol of the DASL-HiCaP trial of darolutamide, along with androgen deprivation therapy, for patients undergoing definitive or salvage radiotherapy. The primary endpoint of this trial is metastasis-free survival (ICECaP-validated), with numerous secondary endpoints including overall survival, prostate cancer-specific survival, PSA-progression-free survival, time to subsequent hormonal therapy, time to castration-resistance, frequency and severity of adverse events, health-related quality of life, fear of recurrence.

| Phase 1 Study of JNJ-69086420, An Actinium-225-Labeled Antibody Targeting Human Kallikrein-2, for Advanced Prostate Cancer |
| Michael J. Morris, MD |
Michael Morris described the rationale, design, and protocol of a phase I trial of 69086420, an actinium-225-labeled antibody targeting human kallikrein-2, for advanced prostate cancer. The primary endpoint of this phase I trial is safety (incidence and severity [grading per NCI-CTCAE V5.0] of AEs including DLTs). Secondary endpoints include prostate-specific antigen response rate, overall response rate (PCWG3 modified RECIST 1.1 criteria), PK, PD, immunogenicity, and biomarker analyses.

| Impact of PSMA PET/CT on Prostate Cancer Salvage Radiotherapy Management: Results from the Prospective Randomized Phase 3 Trial PSMA SRT
|
| Wesley R. Armstrong, BS
|
| Wesley Armstrong discussing results from the prospective randomized phase 3 trial PSMA SRT assessing the impact of PSMA PET/CT on prostate cancer salvage radiotherapy management. In high-risk patients, biochemical recurrence after salvage radiotherapy reaches nearly 50%. Despite improved post-salvage radiotherapy control from RTOG consensus fields and risk-management recommendations, relapse rates remain as high as 50% at 5 years for high-risk patients.

| Progression Patterns by Types of Metastatic Spread, PSA, and Clinical Symptoms: Post-Hoc Analyses of ARAMIS |
| Alicia Morgans, MD, MPH |
In this presentation, Alicia Morgans described the progression patterns of patients with non-metastatic castration resistant prostate cancer (nmCRPC) in the ARAMIS trial. While darolutamide significantly reduced risk of metastatic progression and improved overall survival in men with nmCRPC, it did not change the pattern of metastatic progression. Further, importantly, many patients with nmCRPC experienced metastatic progression without PSA progression, and pain progression was rare.

| Safety and Survival Outcomes in Patients With mCRPC Treated With 177Lu-PSMA After Radium-223: Interim Analysis of the RALU Study |
| Kambiz Rahbar, MD |
| Kambiz Rahbar discussed sequential treatment with lutetium-177–prostate-specific membrane antigen (177Lu-PSMA) after radium-223 (223Ra) in men with metastatic castration-resistant prostate cancer (mCRPC). These real world data demonstrate the feasibility of sequential treatment with 223Ra and 177Lu-PSMA in selected patients with advanced mCRPC. |

| Longitudinal Screening for Depression and Anxiety in Prostate Cancer and Association With Disease and Treatment Factors |
| Risa L. Wong, MD |
| Risa Wong discussed mental health outcomes and screening in men with prostate cancer. Longitudinal screening for depression and anxiety in men with prostate cancer not infrequently identifies men who initially screen negative and subsequently develop significant mental health concerns. Symptoms are associated with ADT and disease or treatment changes, which may inform optimal screening practices. |
