| Lutetium-177-PSMA Therapy in Patients with Prior Radium-223: Safety and Effectiveness Outcomes in the RALU Study |

| In this presentation, Kambiz Rahbar discusses safety and effectiveness outcomes of 177Lu-PSMA therapy in patients with prior treatment with radium-223 in the RALU study. In patients for whom radium-223 had been used as part of routine disease management, subsequent 177Lu-PSMA therapy was clinically feasible and well tolerated, with acceptable myelosuppression rates. Survival outcomes in patients who received the radium-223/177Lu-PSMA sequence were similar to those reported in previous real-world studies and the phase 3 VISION trial. |

Dr. Sartor notes that bone health agents continue to be underutilized. Denosumab cessation is associated with rebound risk, and overuse of skeletal- related event agents persists in castration-sensitive prostate cancer.

| Improvements in Symptoms Related to Bone Metastasis in Recipients of Lutetium-177 PSMA-617 for Prostate Cancer |

| Brian Gonzalez presents work on patient-reported quality of life metrics among men receiving PSMA radioligand therapy (RLT) for the treatment of metastatic castration-resistant prostate cancer (mCRPC). PSMA-RLT conjugates a radionuclide to a small molecule ligand with affinity for PSMA. PSMA-RLT utilizing Lutetium-617 has recently been demonstrated to improve progression-free survival, overall survival, and patient-reported outcomes in the TheraP and VISION trials. However, there is a relative dearth of knowledge about the effects of PSMA-RLT on symptoms related to bone metastasis, a common cause of significant morbidity in men with mCRPC. |

| Guilhem Roubaud presents an analysis of bone mineral density in men treated on the PEACE-1 trial. In terms of assessing the toxicity of abiraterone acetate and prednisone, a planned study amendment sought to analyze whether the addition of abiraterone acetate and prednisone was associated with increased bone loss. It is well known that ADT is associated with bone loss and fracture. However, the involvement of next-generation hormone therapies in this process is somewhat less clear. |
