| Enzalutamide is an FDA approved androgen receptor antagonist used in the treatment of essentially every phase of advanced prostate cancer, from metastatic hormone-sensitive prostate cancer to metastatic and non-metastatic castration-resistant prostate cancer. The approval of enzalutamide for non-metastatic castration-resistant prostate cancer (nmCRPC) was based on the PROSPER trial.

| There is evidence that ADT increases the risk of adverse cardiovascular events such as myocardial infarction, stroke, and cardiovascular mortality, possibly by exacerbating cardiovascular risk factors. In this presentation, Dr. Shore provides data that shows that cardiovascular disease is now the leading cause of mortality in men with prostate cancer.

| In this double-blind, phase 3 trial, men with nonmetastatic, castration-resistant prostate cancer (defined on the basis of conventional imaging and a PSA doubling time of ≤10 months) who were continuing to receive androgen-deprivation therapy were randomly assigned (in a 2:1 ratio) to receive enzalutamide at a dose of 160 mg or placebo once daily.

| Oral Relugolix for Androgen-Deprivation Therapy in Advanced Prostate Cancer

| In this phase 3 trial, the authors randomly assigned patients with advanced prostate cancer, in a 2:1 ratio, to receive relugolix (120 mg orally once daily) or leuprolide (injections every 3 months) for 48 weeks. The primary end point was sustained testosterone suppression to castrate levels (<50 ng per deciliter) through 48 weeks.
